概要: Genetic variations in the root traits of rice remain unexplored in relation to varietal groups and diverse ecosystems. Therefore, we evaluated 257 Bangladeshi accessions, which included landraces and improved varieties belonging to four ecotypes (Aus, Aman, Boro and Jhum), to understand genetic variations in the total root length (TRL), maximum root length and root number under different concentrations of ammonium and nitrate nitrogen in hydroponic culture. There were large variations in all traits across all nitrogen levels. Active elongation of individual roots of the landrace type across all nitrogen conditions suggested a higher potential for the uptake of nutrients by extending roots distribution in any nitrogen environment, while the active development of primary root primordia in the improved type in the presence of nitrate suggested a higher potential for the uptake of nitrate from the soil surface by extending roots distribution around soil surface. Cluster analysis classified the examined accessions into two groups according to their TRL: cluster I with a longer TRL and cluster II with a shorter TRL across all nitrogen conditions. Each of these groups was further divided into two subclusters: sub-cluster Ia, in which the longer TRL resulted from active development of primary root primordia, sub-cluster Ib, in which the longer TRL resulted from active elongation of individual roots, subcluster IIa, in which the shorter TRL resulted from severe reduction of the elongation of individual roots, and sub-cluster IIb, in which the shorter TRL resulted from a moderate reduction in the elongation of individual roots. Further investigation revealed a difference in the composition of these four sub-clusters among the four ecotypes, indicating that Aman was the most diverse ecotype, followed by Aus. The root traits of these promising accessions could be exploited in the future to understand genetic and physiological mechanism(s) of adaptation to diverse ecosystems. 続きを見る

概要: Treatment with tocilizumab (TCZ) to block interleukin-6 (IL-6) signalling is predicted to mitigate cytokine release synd rome (CRS) caused by coronavirus disease 2019 (COVID-19). However, the adverse effects of TCZ on patients with COVID-19 remain unclear. We herein report a patient with COVID-19 treated with TCZ who developed acute hypertriglyceridaemia. Despite favipiravir treatment, acute respiratory distress syndrome developed in a 45-year-old patient with COVID-19; thus, TCZ was initiated. The triglyceride levels greatly increased after TCZ administration. Physicians should consider the negative impact of TCZ on the lipid profile in patients with COVID-19, although COVID-19-induced CRS itself may be an aggravating factor.

概要: This work introduces multi-objective water cycle algorithm (MOWCA) to find the accurate location and size of distributed energy resource (DERs) considering different load models for two seasons (winter, and summer). The impact of uncertainties produced from load and renewable energy resource (RES) such as wind turbine (WT) and photovoltaic (PV) on the performance of the radial distribution system (RDS) are covered as this is closer to the real operation condition. The point estimate method (PEM) is applied for modeling the RES uncertainties. An optimization technique is implemented to find the multi-objective optimal allocation of RESs in RDSs considering uncertainty effect. The main objectives of the work are to maximize the technical, economic and environmental benefits by minimizing different objective functions such as the dissipated power, the voltage deviation, DG cost and total emissions. The proposed multi-objective model is solved by using multi-objective water cycle algorithm (MOWCA), considering the Pareto criterion with nonlinear sorting based on fuzzy mechanism. The proposed algorithm is carried out on different IEEE power systems with various cases.

概要: Background: Primary sclerosing cholangitis (PSC) is a well-known complication of ulcerative colitis (UC), but it is rare to encounter patients requiring both living donor liver transplantation (LDLT) and proctocolectomy. We report a case of elective two-stage surgery involving proctocolectomy performed after LDLT for a patient with early colon cancer concurrent with PSC-related UC. To our knowledge, this is the first report of concurrent cancer successfully treated with both LDLT and proctocolectomy. Case presentation: A 32-year-old Japanese man with colon cancer associated with UC underwent restorative proctocolectomy at 3 months after living donor liver transplantation (LDLT) for PSC. He was diagnosed with PSC and UC when he was a teenager. Conservative therapy was initiated to treat both PSC and UC. He had experienced recurrent cholangitis for years; therefore, a biliary stent was placed endoscopically. However, his liver function progressively deteriorated. Colonoscopic surveillance revealed early colon cancer; hence, surgical treatment was considered. PSC progressed to cirrhosis and portal hypertension; hence, LDLT was performed before restorative proctocolectomy. Three months after LDLT, we performed restorative proctocolectomy with ileal pouch–anal anastomosis. The postoperative course was uneventful. The patient was well, with good liver and bowel functions and without tumor recurrence, more than 1 year after proctocolectomy. Conclusions: With strict patient selection and careful patient management and follow-up, elective proctocolectomy may be performed safely and effectively after LDLT for concurrent early colon cancer with PSC-related UC. There are no previous reports of the use of both LDLT and proctocolectomy for the successful treatment of PSC-related UC and concurrent cancer. 続きを見る

概要: Dilated cardiomyopathy (DCM) is a fatal heart disease characterized by left ventricular dilatation and cardiac dysfuncti on. Recent genetic studies on DCM have identified causative mutations in over 60 genes, including RBM20, which encodes a regulator of heart-specific splicing. DCM patients with RBM20 mutations have been reported to present with more severe cardiac phenotypes, including impaired cardiac function, atrial fibrillation (AF), and ventricular arrhythmias leading to sudden cardiac death, compared to those with mutations in the other genes. An RSRSP stretch of RBM20, a hotspot of missense mutations found in patients with idiopathic DCM, functions as a crucial part of its nuclear localization signals. However, the relationship between mutations in the RSRSP stretch and cardiac phenotypes has never been assessed in an animal model. Here, we show that Rbm20 mutant mice harboring a missense mutation S637A in the RSRSP stretch, mimicking that in a DCM patient, demonstrated severe cardiac dysfunction and spontaneous AF and ventricular arrhythmias mimicking the clinical state in patients. In contrast, Rbm20 mutant mice with frame-shifting deletion demonstrated less severe phenotypes, although loss of RBM20-dependent alternative splicing was indistinguishable. RBM20^ protein cannot be localized to the nuclear speckles, but accumulated in cytoplasmic, perinuclear granule-like structures in cardiomyocytes, which might contribute to the more severe cardiac phenotypes.

概要: “One-pot regeneration”, which is simple regneneration method of ammonia borane (AB) using hydrazine and liquid ammonia, enables conversion of AB from hexagonal boron nitride (h-BN) after milling hydrogenation. Solution ^<11>B-NMR revealed the presence of AB after NH_3/N_2H_4 treatment of milled h-BN (BNH_x) although the yield of AB was less than 5%. The conversion mechanism was clarified as B-H bonds on the h-BN surface created by ball-milling under hydrogen pressure have an ability to form AB, which was confirmed by Thermogravimetry-Residual Gas Analysis (TG-RGA) and Infrared (IR) analysis. The reaction routes are also the same as regeneration route of polyborazylene because intermediates of AB such as (B(NH_2)_3 and hydrazine borane were found by solution ^<11>B-NMR after soaking BNH_x in liquid NH_3 and hydrazine, respectively. Because of the fact that all reactions proceed on the h-BN surface and no reaction proceeds when neat h-BN is treated, breaking of B_3N_3 ring structure and then creation of B-H bond is the key issue to increase conversion yield of AB.

概要: A bizarre crab of the family Leucosiidae, Oreotlos pala Tan & Ng, 1996 is redescribed on the basis of four specimens, in cluding three males and one female, collected from the Ryukyu Islands. This species was known only by the female holotype from Guam, the Mariana Islands. Our material from the Ryukyus enable us to describe the morphology of taxonomically important male characters (abdomen, telson, and gonopods) of this poorly known species for the first time. The assignment of the species to Oreotlos is fully confirmed.
久米島および宮古群島の伊良部島より採集された4個体の標本に基づき,コブシガニ科ツノカルイシコブシ属Oreotlos Ihle, 1918 のOreotlos pala Tan & Ng, 1996を再記載した.本種は,マリアナ諸島グアム島で採集された雌1個体に基づいて記載され,その後,川本・奥野(2003)による一般向けのガイドブックに久米島で撮影された個体の水中写真が掲載されているにすぎない稀種であった.本種の雄の形態はこれまで知られておらず,またホロタイプは鉗脚と歩脚を欠いているが,本研究の検討標本は原記載にも示されている頭胸甲背面の肝域後方,側方突出部,鰓域後方にそれぞれ1個大きく深いくぼみを有することにより,同種であると判断した.本種には標準和名が与えられていなかったため,頭胸甲背面に月面のクレーターを彷佛させる顕著なくぼみを有することから,ゲツメンカルイシコブシの新称を提唱する.

概要: For simultaneous screening of mixed features (MF) by DSM-5 and mixed depression (MD) by Benazzi, useful symptoms were ex tracted from our 12-item dimensional scale for depressive mixed state (DMX-12). Subjects were 190 consecutive cases with major depressive episode (MDE) who visited our clinic. Associations between symptomatological combinations of the DMX-12 and MF or MD were analyzed using receiver operating characteristic (ROC). The rate of MF was 4.2% while that of MD was 22.6%. Eight symptoms (overreactivity, inner tension, racing/crowded thought, impulsivity, irritability, aggression, risk-taking behavior, and dysphoria) with their AUC > 0.6 for ROC curves were specially focused on distinguishing patients with MF or MD from non-mixed patients. By using these 8 symptoms, 40.5% of the overall patients were screened as positive at the same cut-off value (≥13) for both MD and MF. The AUC of ROC curve and sensitivity/specificity were well balanced together with sufficient negative predictive values. The abovementioned 8 symptoms seem to be helpful for primary screening and negative check of DMX with considerable severity during MDE.

概要: Studies have suggested that fishes with broad distribution ranges, long pelagic duration, and great mobility in their ea rly life histories may have an advantage in their ability to return to their natal reef habitats. Siganus spinus is a siganid fish with a broad dispersal strategy, which may also exploit its great mobility to enhance self-recruitment. Variations in mitochondrial DNA sequences were analysed to estimate the genetic structure and demographic history of Siganus spinus in the West Pacific. In total, 446 nucleotide sequences, including the tRNAThr gene, the tRNAPro gene, and the first half of the control region, were analysed from 240 individuals sampled at five sites spanning the West Pacific Ocean; 37 haplotypes were identified. Our results showed significant geographic genetic structure as well as isolation by distance. Both neutrality tests and mismatch distribution indicated that the population expansion of S. spinus through the study area may have occurred in the late Quaternary. A dispersal strategy characterised by large juveniles and open-sea distribution may have contributed to a sudden population expansion after the last glacial period, along with an expansion of distribution.

概要: CeTe_3 is a unique platform to investigate the itinerant magnetism in a van der Waals (vdW) coupled metal. Despite chemi cal pressure being a promising route to boost quantum fluctuation in this system, a systematic study on the chemical pressure effect on Ce^<3+>(4f^1) states is absent. Here, we report on the successful growth of a series of Se doped single crystals of CeTe_3. We found a fluctuation driven exotic magnetic rotation from the usual easy-axis ordering to an unusual hard-axis ordering. Unlike in localized magnetic systems, near-critical magnetism can increase itinerancy hand-in-hand with enhancing fluctuation of magnetism. Thus, seemingly unstable hard-axis ordering emerges through kinetic energy gain, with the self-consistent observation of enhanced magnetic fluctuation (disorder). As far as we recognize, this order-by-disorder process in fermionic system is observed for the first time within vdW materials. Our finding opens a unique experimental platform for direct visualization of the rich quasiparticle Fermi surface deformation associated with the Fermionic order-by-disorder process. Also, the search for emergent exotic phases by further tuning of quantum fluctuation is suggested as a promising future challenge.

概要: Urban trees are under unprecedented pressure and competition worldwide with other land uses. Homestead windbreaks in urb an areas are an important part of urban forests because of their proximity to settlements. To aid in the conservation of old-growth homestead trees in the urban setting on Ryukyu Archipelago, Japan, this article surveys the dimensions and spatial distribution of century-old trees planted as windbreaks along homestead borderlines on Ishigaki Island in the Ryukyu Archipelago and the historical change caused by urban sprawl. The homesteads studied in this article do not match the scale of a traditional Western context and usually consist of an area of less than 200 square meters on Okinawa. A combined approach consisting of field surveys and the study of aerial photos was applied to identify changes in spatial distribution of tree lines surrounding the houses in 1945, 1972 and the present. We measured the dimensions of 1659 Fukugi trees with a minimum diameter at breast height (DBH) of 5 cm. The mean tree height, mean DBH and mean estimated tree age were 7.3 m, 26.9 cm and 107.5 years, respectively. Homestead trees are not only useful as windbreaks, timber sources and musical instruments, but have also been historically resilient in the face of strong typhoons and catastrophic tsunamis in the region. Over 60% of the surveyed trees were planted on the east and north sides of homesteads as protection from typhoons and monsoonal winds in the winter. In addition to G. subelliptica, other tree species, namely Podocarpus macrophyllus and Diospyros egbert-walkeri, have been commonly used as homestead windbreaks. However, homestead windbreaks in highly urbanized regions are generally declining and have experienced fragmentation, lower tree density and shorter tree height than those in rural areas. Because of the small number of trees older than 200 years, we assume that high urbanization has jeopardized old-growth trees. The demand for settlement land dramatically increases as the population increases and household-size decreases, creating more households. Therefore, a conservation project involving multiple stakeholders must be developed to conserve old-growth trees in urban settings.

概要: Wide variations of typhoon-induced spikelet sterility and days to heading (DH) were found among 317 introgression lines (INLs) originating from new-plant-type varieties with a genetic background of Indica Group rice variety IR 64. Sterility was induced by continuous dry winds from the typhoon. The most seriously affected were 194 INLs with DH ranging from 83 to 86 days. Positive correlations with the spikelet sterility score (SSS) were found for culm length (CL), panicle length (PL), and panicle weight (PW). Based on these traits and DH, the 194 INLs were classified into three groups: A1, A2, and B. The means of SSS and four traits (CL, PL, culm and leaf weight (CW), and panicle weight (PW)) were the lowest in group A1, and highest in group B. PN was significantly higher in group B than those in the others. Seven quantitative trait loci (QTL) for SSS were detected on chromosomes (chr.) 1, 2, 4, 7 (two), 8, and 11. Among them, the QTL of chr. 4 was located in the same regions as those of DH, CL, PL, CW, and panicle number (PN). 概要: The fiddler crab, Austruca lactea inhabits tidal flat areas and is widely distributed across Japan, the Korean Peninsula , China, Taiwan and northern Vietnam. Fiddler crab habitat is being lost rapidly due to human impacts and this species is currently listed as endangered in Japan. We studied the population genetic structure of A. lactea using mitochondrial DNA control region markers to define management units. A. lactea individuals were sampled from Osaka, Hiroshima and Kumamoto in Japan, and from Taichung and the Penghu Islands in Taiwan. All local samples exhibited high levels of genetic diversity, and no sampled populations showed evidence for a significant decline in effective population size that can result from population bottleneck effects. Pairwise FST estimates distinguished three discrete A. lactea populations corresponding with, the Seto Inland Sea (Osaka and Hiroshima), Kyushu (Kumamoto) and Taiwan. The three populations showed clear differences in historical population expansion times and their population dynamics after expansion. Results of the study indicate that A. lactea dispersal is limited geographically and that high levels of genetic diversity are maintained both within and among populations.

概要: Background: The aim of this study was to determine the effectiveness of the Smart Life Stay (SLS) program, which is an e xperience-oriented stayover program, in combination with health tourism and mandatory health guidance on glucose metabolism after 2 years. Methods: The participants of the SLS program (n = 792) were recruited from a database of 23 medical insurers. They underwent a mandatory health examination termed Specific Health Checkups in 2014. The participants were included if they had diabetes or were at a high risk of diabetes and if they satisfied the following inclusion criteria: (1) body mass index (BMI; kg/m^2 ) > 25, or (2) waist circumference (WC; cm) > 85 for men and > 90 for women, or (3) hemoglobin A1c (HbA1c; %) > 5.6, or (4) fasting plasma glucose (FPG; mg/dl) > 100. Individuals who corresponded to one or more items were included as study participants. The control subjects (n = 3645) were nonparticipants of the program who were selected from the database and met the inclusion criteria. The lifestyle changes and changes in mean BMI, WC, FPG, and HbA1c in both groups from baseline to 2-year follow-up were compared by inverse probability weighting of a propensity score. Results: The percentage of people who exercised regularly increased significantly in the SLS group compared with the control group. In the SLS group, BW, BMI, and WC significantly decreased by 1.75 kg, 0.60 kg/m^2 , and 1.45 cm, respectively, whereas in the control group, WC, FPG, and HbA1c increased significantly by 0.38 cm, 3.37 mg/dl, and 0.12%, respectively. The comparison between groups revealed that the BW, BMI, WC, FPG, and HbA1c improved significantly in the SLS group. Conclusions: The SLS program is suggested to help improve glucose metabolism. This program could be a feasible option as a lifestyle intervention program for diabetes.

概要: Oncoprotein E6 of high-risk human papillomavirus (HPV) plays a critical role in inducing cell immortalization and malign ancy. E6 downregulates caspase-dependent pathway through the degradation of p53. However, the effect of HPV E6 on other pathways is still under investigation. In the present study, we found that HPV E6 directly binds to all three forms (precursor, mature, and apoptotic) of apoptosis-inducing factor (AIF) and co-localizes with apoptotic AIF. This binding induced MG132-sensitive reduction of AIF expression in the presence of E6 derived from HPV16 (16E6), a cancer-causing type of HPV. Conversely, E6 derived from a non-cancer-causing type of HPV, HPV6 (6E6), did not reduce the levels of AIF despite its interaction with AIF. Flow cytometric analysis revealed that 16E6, but not 6E6, suppressed apoptotic AIF-induced chromatin degradation (an indicator of caspase-independent apoptosis) and staurosporine (STS, a protein kinase inhibitor)-induced apoptosis. AIF knockdown reduced STS-induced apoptosis in both of 16E6-expressing and 6E6-expressing cells; however, the reduction in 16E6-expressing cells was lower than that in 6E6-expressing cells. These findings indicate that 16E6, but not 6E6, blocks AIF-mediated apoptosis, and that AIF may represent a novel therapeutic target for HPV-induced cervical cancer.

Ancient genomes at the edge of the region may elucidate a more detailed picture of the peopling of East Eurasia. Here, we analyze the whole-genome sequence of a 2,500-year-old individual (IK002) from the main-island of Japan that is characterized with a typical Jomon culture. The phylogenetic analyses support multiple waves of migration, with IK002 forming a basal lineage to the East and Northeast Asian genomes examined, likely representing some of the earliest-wave migrants who went north from Southeast Asia to East Asia. Furthermore, IK002 shows strong genetic affinity with the indigenous Taiwan aborigines, which may support a coastal route of the Jomon-ancestry migration. This study highlights the power of ancient genomics to provide new insights into the complex history of human migration into East Eurasia.

概要: The radiation doses from natural radiation sources in Japan are reviewed using the latest knowledge. The United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) and the Nuclear Safety Research Association report the annual effective doses from cosmic rays, terrestrial radiation, inhalation, and ingestion as natural sources. In this paper, the total annual effective dose from cosmic-ray exposure is evaluated as 0.29 mSv. The arithmetic mean of the annual effective dose from external exposure to terrestrial radiation is 0.33 mSv for the Japanese population using the data of nationwide surveys by the National Institute of Radiological Sciences. Previously in Japan, although three different groups have conducted nationwide indoor radon surveys using passive-type radon monitors, to date only the Japan Chemical Analysis Center (JCAC) has performed a nationwide radon survey using a unified method for radon measurements conducted indoor, outdoor, and in the workplace. Consequently, the JCAC results are used for the annual effective dose from radon and that for radon inhalation is estimated as 0.50 mSv using a current dose conversion factor. In this paper, UNSCEAR values are used for the mean indoor and outdoor thoron-progeny concentrations, and the annual effective dose from thoron is reported as 0.09 mSv. Thus, the annual effective dose from radon and thoron inhalation is 0.59 mSv. From a JCAC large-scale survey of foodstuffs, the committed effective dose from the main radionuclides in dietary intake is 0.99 mSv. Finally, the Japanese population dose from natural radiation is given as 2.2 mSv, which is similar to the reported global average of 2.4 mSv.

概要: Background:Leptospirosis is one of the most significant zoonoses across the world not only because of its impact on huma n and animal health but also because of the economic and social impact on agrarian communities. Leptospirosis is endemic in Sri Lanka where paddy farming activities, the use of draught animals in agriculture, and peridomestic animals in urban and rural areas play important roles in maintaining the infection cycle of pathogenic Leptospira, especially concerning animals as a potential reservoir. In this study, an environmental DNA (eDNA) metabarcoding methodology was applied in two different agro-ecological regions of Sri Lanka to understand the eco-epidemiology of leptospirosis.\nMethodology/Principal findings:Irrigation water samples were collected in Kandy District (wet zone mid-country region 2) and Girandurukotte, Badulla District (intermediate zone low-country region 2); and analysed for the presence of pathogenic Leptospira, associated microbiome and the potential reservoir animals. Briefly, we generated PCR products for high-throughput sequencing of multiple amplicons through next-generation sequencing. The analysis of eDNA showed different environmental microbiomes in both regions and a higher diversity of Leptospira species circulating in Kandy than in Girandurukotte. Moreover, the number of sequence reads of pathogenic Leptospira species associated with clinical cases such as L. interrogans was higher in Kandy than in Girandurukotte. Kandy also showed more animal species associated with pathogenic bacterial species than Girandurukotte. Finally, several pathogenic bacterial species including Arcobacter cryaerophilus, responsible for abortion in animals, was shown to be associated with pathogenic Leptospira.\nConclusions/Significance:Leptospirosis has been considered to be endemic in wet regions, consistently, leptospiral sequences were detected strongly in Kandy. The great Leptospira species diversity in Kandy observed in this study shows that the etiological agents of leptospirosis in Sri Lanka might be underestimated. Furthermore, our eDNA metabarcoding can be used to discriminate bacterial and animal species diversity in different regions and to explore environmental microbiomes to identify other associated bacterial pathogens in the environment.

概要: Serum markers that differentiate between tuberculous and non-tuberculous pneumonia would be clinically useful. However, few serum markers have been investigated for their association with either disease. In this study, serum levels of interferon gamma (IFN-γ), matrix metalloproteinases 1 and 9 (MMP-1 and MMP-9, respectively), and periostin were compared between 40 pulmonary tuberculosis (PTB) and 28 non-tuberculous pneumonia (non-PTB) patients. Diagnostic performance was assessed by analysis of receiver-operating characteristic (ROC) curves and classification trees. Serum IFN-γ and MMP-1 levels were significantly higher and serum MMP-9 levels significantly lower in PTB than in non- PTB patients (p < 0.001, p = 0.002, p < 0.001, respectively). No significant difference was observed in serum periostin levels between groups. ROC curve analysis could not determine the appropriate cut-off value with high sensitivity and specificity; therefore, a classification tree method was applied. This method identified patients with limited infiltration into three groups with statistical significance (p = 0.01), and those with MMP-1 levels < 0.01 ng/ mL and periostin levels ≥ 118.8 ng/mL included only non-PTB patients (95% confidence interval 0.0–41.0). Patients with extensive infiltration were also divided into three groups with statistical significance (p < 0.001), and those with MMP-9 levels < 3.009 ng/mL included only PTB patients (95% confidence interval 76.8–100.0). In conclusion, the novel classification tree developed using MMP-1, MMP-9, and periostin data distinguished PTB from non- PTB patients. Further studies are needed to validate our cut-off values and the overall clinical usefulness of these markers.

概要: Background:To treat small-for-size syndrome (SFSS) after living donor liver transplantation (LDLT), many procedures were described for portal flow modulation before, during, or after transplantation. The selection of the procedure as well as the best timing remains controversial.\nCase presentation:A 43-year-old female with end-stage liver disease underwent LDLT with extended left with caudate lobe graft from her donor who was her 41-year-old brother (graft volume/standard liver volume (GV/SLV), 35.7%; graft to recipient weight ratio (GRWR), 0.67%). During the surgery, splenectomy could not be performed owing to severe peri-splenic adhesions to avoid the ruined bleedings. The splenic artery ligation was not also completely done because it was dorsal to the pancreas and difficult to be approached. Finally, adequate portal vein (PV) inflow was confirmed after portal venous thrombectomy. As having post-transplant optional procedures that are accessible for PV flow modulation, any other procedures for PV modulation during LDLT were not done until the postoperative assessment of the graft function and PV flow for possible postoperative modulation of the portal flow accordingly. Postoperative PV flow kept as high as 30 cm/s. By the end of the 1st week, there was a progressive deterioration of the total bilirubin profile (peak as 19.4 mg/dL) and ascitic fluid amount exceeded 1000 mL/day. Therefore, splenic artery embolization was done effectively and safely on the 10th postoperative day (POD) to reverse early allograft dysfunction as PV flow significantly decreased to keep within 20 cm/s and serum total bilirubin levels gradually declined with decreased amounts of ascites below 500 mL on POD 11 and thereafter. The patient was discharged on POD 28 with good condition.\nConclusions:SFSS can be prevented or reversed by the portal inflow modulation, even by post-transplant procedure. This case emphasizes that keeping accessible angiographic treatment options for PV modulation, such as splenic artery embolization, after LDLT is quite feasible.

概要: Background: Guideline for Pharmacological Therapy for Schizophrenia was published by the Japanese Society of Neuropsycho pharmacology in 2015. “Effectiveness of Guidelines for Dissemination and Education in psychiatric treatment (EGUIDE)” project aimed to standardize medical practice using quality indicators (QIs) as indices to evaluate the quality of medical practice. In this study, we have reported the quality indicator values of prescription before the beginning of the guideline lectures in the EGUIDE project to ascertain the baseline status of treating patients with schizophrenia. Methods: A cross-sectional, retrospective case record survey was conducted, involving 1164 patients with schizophrenia at the time of discharge. We checked all types and dosage of psychotropic drugs. Results: Forty-three percent of patients had antipsychotic polypharmacy, and substantial concomitant medication was observed (antidepressants; 8%, mood stabilizers: 37%, anxiolytics or hypnotics: 68%). Conclusions: In the results obtained in this study, we plant to report changes in the effectiveness of education in the EGUIDE project near the future. 続きを見る

However, CRISPR–Cas9 is rarely used to analyze promoter regions. The insulin promoter region (approximately 1,000 bp) directs β cell-specific expression of insulin, which in vitro studies show is regulated by ubiquitous, as well as pancreatic, β cell-specific transcription factors. However, we are unaware of any confirmatory in vivo studies. Here, we used CRISPR–Cas9 technology to generate mice with mutations in the promoter regions of the insulin I (Ins1) and II (Ins2) genes. We generated 4 homozygous diabetic mice with 2 distinct mutations in the highly conserved C1 elements in each of the Ins1 and Ins2 promoters (3 deletions and 1 replacement in total). Remarkably, all mice with homozygous or heterozygous mutations in other loci were not diabetic. Thus, the C1 element in mice is required for Ins transcription in vivo.

概要: Introduction The speed of declining kidney function differs among patients with diabetic nephropathy. This study was und ertaken to clarify clinical and pathological features that affect the speed of declining kidney function in patients with diabetic nephropathy.\nResearch design and methods This study was design as multicenter retrospective study. The subjects (377 patients with diabetic nephropathy diagnosed by kidney biopsy at 13 centers in Japan) were classified into three groups based on the estimated glomerular filtration rate (eGFR) declining speed. The eGFR increasing group, the control group, and the eGFR declining group were divided at 0 and 5 mL/min/1.73 m2/year, respectively. Characteristics of clinicopathological findings of declining kidney function were evaluated.\nResults The mean observation period of this study was 6.9 years. The control group, the eGFR increasing group, and the eGFR declining group included 81, 66, and 230 patients, respectively. The incidences of composite kidney events represented by 100 persons/year were 25.8 in the eGFR declining group and 2.0 in the eGFR increasing group. After adjustment for age, sex, systolic blood pressure, hemoglobin, and urinary albumin levels, three clinicopathological findings (urinary albumin levels, presence of nodular lesion, and mesangiolysis) were risk factors for inclusion in the eGFR declining group (the ORs were 1.49, 2.18, and 2.08, respectively). In contrast, the presence of subendothelial space widening and polar vasculosis were characteristic findings for inclusion in the eGFR increasing group (the ORs were 0.53 and 0.41, respectively).\nConclusions As well as urinary albumin elevation, nodular lesion and mesangiolysis were characteristic pathological features of patients with fast declining kidney function.

概要: In plants, growth–defense tradeofs are essential for optimizing plant performance and adaptation under stress conditions , such as pathogen attack. Root-knot nematodes (RKNs) cause severe economic losses in many crops worldwide, although little is known about the mechanisms that control plant growth and defense responses during nematode attack. Upon investigation of Arabidopsis thaliana infected with RKN (Meloidogyne incognita), we observed that the atypical transcription factor DP-E2F-like 1 (DEL1) repressed salicylic acid (SA) accumulation in RKN-induced galls. The DEL1-defcient Arabidopsis mutant (del1-1) exhibited excessive SA accumulation in galls and is more resistant to RKN infection. In addition, excessive lignifcation was observed in galls of del1-1. On the other hand, the root growth of del1-1 is reduced after RKN infection. Taken together, these fndings suggest that DEL1 plays an important role in the balance between plant growth and defense responses to RKN infection by controlling SA accumulation and lignifcation. 続きを見る

概要: We compare Bi_2Te_3 powders prepared by conventional ball milling to powders milled in supercritical carbon dioxide (scC O_2). We demonstrate that scCO_2 milling overcomes size-reduction limitations reported for conventional milling. XRD and TEM reveal nanograins with smaller average sizes (< 10 nm) and narrower grain size distributions in the scCO_2 milled case. scCO_2 milling also preserves the crystallinity and shows less oxidation than conventional milling. This is the first report of Bi_2Te_3 with a sub-10 nm grain size whilst conserving high quality crystallinity, made using a top-down approach. Our study offers a route for developing unprecedentedly fine bulk nanostructured Bi_2Te_3-based thermoelectric materials.

概要: This paper presents an evaluation of a rapid pretreatment method for tritium analysis, that uses ion exchange resins (th e batch method). The standard water samples were prepared with river water collected in Okinawa, Japan. Powdex resin was used in this study, which is an ion exchange resins in the form of powder beads. First, 100 mL of the standard sample was stirred with the cation exchange (PCH) and anion exchange (PAO) resins, as well as with activated carbon. To determine the optimal stirring time, we tested stirring times of 5, 10, 20 and 30 min. Additionally, to determine a suitable amount of ion exchange resin for environmental water samples, variations in the amount of resin and activated carbon were tested. Under the conditions of Experiment-3 (0.3 g, 0.3 g and 0.6 g of PCH, PAO and activated carbon, respectively) and with a stirring time of 5 min, electrical conductivity values of less than 1 mSm^<−1> were achieved. These results demonstrate the effectiveness of the removal of impurities in the sample water by ion exchange resins and confirm that such an effect can be achieved during a short period of time (i.e., in 5 min).

概要: Idiopathic nephrotic syndrome newly affects 1–3 per 100,000 children per year. Approximately 85% of cases show complete remission of proteinuria following glucocorticoid treatment. Patients who do not achieve complete remission within 4–6 weeks of glucocorticoid treatment have steroid-resistant nephrotic syndrome (SRNS). In 10–30% of steroid-resistant patients, mutations in podocyte-associated genes can be detected, whereas an undefined circulating factor of immune origin is assumed in the remaining ones. Diagnosis and management of SRNS is a great challenge due to its heterogeneous etiology, frequent lack of remission by further immunosuppressive treatment, and severe complications including the development of end-stage kidney disease and recurrence after renal transplantation. A team of experts including pediatric nephrologists and renal geneticists from the International Pediatric Nephrology Association (IPNA), a renal pathologist, and an adult nephrologist have now developed comprehensive clinical practice recommendations on the diagnosis and management of SRNS in children. The team performed a systematic literature review on 9 clinically relevant PICO (Patient or Population covered, Intervention, Comparator, Outcome) questions, formulated recommendations and formally graded them at a consensus meeting, with input from patient representatives and a dietician acting as external advisors and a voting panel of pediatric nephrologists. Research recommendations are also given.

概要: Kaposi’s sarcoma-associated herpesvirus (KSHV) causes both AIDS-related Kaposi’s sarcoma (KS) and classic KS, but their clinical presentations are different, and respective mechanisms remain to be elucidated. The KSHV K1 gene is reportedly involved in tumorigenesis through the immunoreceptor tyrosine-based activation motif (ITAM). Since we found the sequence variations in the K1 gene of KSHV isolated from AIDS-related KS and classic KS, we hypothesized that the transformation activity of the K1 gene contributes to the different clinical presentations. To evaluate our hypothesis, we compared the transformation activities of the K1 gene between AIDS-related KS and classic KS. We also analyzed ITAM activities and the downstream AKT and NF-κB. We found that the transformation activity of AIDS-related K1 was greater than that of classic K1, and that AIDS-related K1 induced higher ITAM activity than classic K1, causing more potent Akt and NF-κB activities. K1 downregulation by siRNA in AIDS-related K1 expressing cells induced a loss of transformation properties and decreased both Akt and NF-κB activities, suggesting a correlation between the transformation activity of K1 and ITAM signaling. Our study indicates that the increased transformation activity of AIDS-related K1 is associated with its clinical aggressiveness, whereas the weak transformation activity of classic type K1 is associated with a mild clinical presentation and spontaneous regression. The mechanism of spontaneous regression of classic KS may provide new therapeutic strategy to cancer.

概要: People in the Solomon Islands today are considered to have derived from Asian- and Papuan-related ancestors. Papuan-rela ted ancestors colonized Near Oceania about 47,000 years ago, and Asian-related ancestors were Austronesian (AN)-speaking population, called Lapita, who migrated from Southeast Asia about 3,500 years ago. These two ancestral populations admixed in Near Oceania before the expansion of Lapita people into Remote Oceania. To understand the impact of the admixture on the adaptation of AN-speaking Melanesians in Near Oceania, we performed the genome-wide single nucleotide polymorphism (SNP) analysis of 21 individuals from Munda, the main town of the New Georgia Islands in the western Solomon Islands. Population samples from Munda were genetically similar to other Solomon Island population samples. The analysis of genetic contribution from the two different ancestries to the Munda genome revealed significantly higher proportions of Asian- and Papuan-related ancestries in the region containing the annexin A1 (ANXA1) gene (Asian component > 82.6%) and in the human leukocyte antigen (HLA) class II region (Papuan component > 85.4%), respectively. These regions were suspected to have undergone natural selection since the time of admixture. Our results suggest that admixture had affected adaptation of AN-speaking Melanesians in the Solomon Islands.

概要: Carnitine-acylcarnitine translocase (CACT) deficiency is a fatty acid ß-oxidation disorder of the carnitine shuttle in m itochondria, with a high mortality rate in childhood. We evaluated three patients, including two siblings, with neonatal-onset CACT deficiency and revealed identical homozygous missense mutations of p.Arg275Gln within the SLC25A20 gene. One patient died from hypoglycemia and arrhythmia at 26 months; his pathological autopsy revealed increased and enlarged mitochondria in the heart but not in the liver.

概要: Research Highlights: This study contributes to the improvement of the understanding of ecosystem functions of trees grow ing outside the forest, by quantifying the carbon sequestration function of a homestead windbreak, for example, a linear forest belt planted bordering a farmhouse in small islands. Background and objectives: Carbon storage in small-scale stands of forests have been less studied compared to that in large-scale forests. The aims of the present study were to clarify the ecological functions of carbon storage and the economic value of homestead windbreaks to propose effective conservation strategies for old-growth homestead windbreaks in the face of climate change. Materials and Methods: On the small islands of Okinawa Prefecture, the dominant tree species used for the homestead windbreaks is fukugi (Garcinia subelliptica Merr.). We collected data on the diameter at breast height (DBH) and the height of 23,518 fukugi trees in 10 villages from 2009 to 2018. Results: The total amount of carbon stored in the remnant fukugi homestead trees of the 10 surveyed hamlets was 6089 t-CO_2. The amount is equivalent to the carbon amount that is stored in a 40-year-old Japanese cedar (Cryptomeria japonica) forest, a representative tree species in Japan, of 20.9 ha area. Furthermore, the estimated economic value of the homestead trees was equivalent to USD 235,433, in terms of the plantation and management costs of 40-year-old Japanese cedar forests. This study revealed that homestead trees planted in an orderly line usually have a high density; hence, they have a high potential for biomass accumulation, carbon sequestration, and climate change mitigation. Moreover, homestead trees could contribute to a reduction in carbon diffusion, by cooling the house and reducing potential energy consumption. The findings related to homestead trees are consistent with those of other types of trees outside forests or small patches of trees: not adding to future land use competition and highly effective at carbon sequestration. Conclusions: The finding related to the carbon storage of homestead trees will provide basic information, as well as a new perspective on future local conservation and its contribution to climate change mitigation. This study suggests the necessity of the existing trees being properly managed, recruiting trees to be planted to replace old-growth trees, and replanting trees near newly established houses or old homesteads where trees have been cut.

概要: Both thin basement membrane nephropathy (TBMN) and autosomal dominant Alport syndrome (ADAS) are types of hereditary nep hritis resulting from heterozygous mutations in COL4A3 or COL4A4 genes. Although TBMN is characterized by hematuria and thinning of the glomerular basement membrane (GBM) with excellent renal prognosis, some patients develop end-stage renal disease (ESRD) later in life. In contrast, although AS is characterized by progressive nephropathy with lamellation of the GBM, there are some patients diagnosed with ADAS from a family history of ESRD but who only suffer from hematuria with GBM thinning. These findings indicate a limitation in distinction between TBMN and ADAS. Diagnosis of AS is significant because it facilitates careful follow-up and early treatment, whereas diagnosis of TBMN can underestimate the risk of ESRD. However, some experts are against using the term ADAS as the phenotypes of heterozygous variants vary from no urinary abnormality to ESRD, even between family members with the same mutations, indicating that unknown secondary factors may play a large role in the disease severity. These diagnostic difficulties result in significant confusion in clinical settings. Moreover, recent studies revealed that the number of patients with chronic kidney disease caused by these gene mutations is far higher than previously thought. The aim of this article is to review differing opinions regarding the diagnosis of heterozygous COL4A3 or COL4A4 variants, and to highlight the importance for nephrologists to recognize this disease, and the importance of the need to reclassify this disease to minimize the current confusion.

概要: Adult T-cell leukemia/lymphoma (ATL) is a human T-cell leukemia virus type 1 (HTLV-1)–associated T-cell malignancy with generally poor prognosis. Although only ∼5% of HTLV-1 carriers progress to ATL, early diagnosis is challenging because of the lack of ATL biomarkers. In this study, we analyzed blood plasma profiles of asymptomatic HTLV-1 carriers (ACs); untreated ATL patients, including acute, lymphoma, smoldering, and chronic types; and ATL patients in remission. Through SOMAscan, expression levels of 1305 plasma proteins were analyzed in 85 samples (AC, n = 40; ATL, n = 40; remission, n = 5). Using gene set enrichment analysis and gene ontology, overrepresented pathways in ATL vs AC included angiogenesis, inflammation by cytokines and chemokines, interleukin-6 (IL-6)/JAK/STAT3, and notch signaling. In selecting candidate biomarkers, we focused on soluble tumor necrosis factor receptor 2 (sTNFR2) because of its active role in enriched pathways, extreme significance (Welch’s t test P < .00001), high discrimination capacity (area under the curve >0.90), and novelty in ATL research. Quantification of sTNFR2 in 102 plasma samples (AC, n = 30; ATL, n = 68; remission, n = 4) using enzyme-linked immunosorbent assay showed remarkable elevations in acute ATL, at least 10 times those of AC samples, and return of sTNFR2 to AC state levels after achieving remission. Flow cytometry and immunostaining validated the expression of TNFR2 in ATL cells. No correlation between sIL-2 and sTNFR2 levels in acute ATL was found, suggesting the possibility of sTNFR2 as an independent biomarker. Our findings represent the first extensive blood-based proteomic analysis of ATL, suggesting the potential clinical utility of sTNFR2 in diagnosing acute ATL.

概要: Aims: Dermatopathic reaction is a histopathological finding of lymph nodes that usually occurs in patients with inflammatory pruritic cutaneous lesions. However, it is sometimes seen in patients with cutaneous T cell lymphoma. Adult T cell leukaemia/lymphoma (ATLL) is a T cell malignancy caused by infection with human T cell leukaemia virus type I (HTLV-1), which is frequently accompanied by cutaneous lesions. However, the detailed clinicopathological characteristics of the dermatopathic reaction of lymph nodes in ATLL patients and HTLV-1 carriers, addressed in this study, remains to be clarified. Methods and results: We retrospectively analysed 18 nodal lesions with dermatopathic reaction in HTLV-1 carriers. Axillary and inguinal lymph nodes were the primary affected tissues. Three cases with atypical lymphoid cell infiltration were defined as ATLL with dermatopathic reaction (ATLL-D), showing an abnormal T cell immunophenotype and T cell monoclonality. Two of the three ATLL-D patients died 14 and 7 months after diagnosis (the third case had a very short follow-up). The other 15 patients were indistinguishable from reactive lesions and were defined as HTLV-1-associated lymphadenitis with dermatopathic reaction (HAL-D). They showed an indolent clinical course, with only one case eventually transforming to aggressive disease. Conclusions: Lymph node lesions accompanied by dermatopathic reaction in HTLV1 carriers represent a spectrum that includes reactive and neoplastic conditions. HAL-D should be distinguished from ATLL-D, especially to avoid overtreatment. 続きを見る

概要: The occurrence of a little known upogebiid mud shrimp, Upogebia pugnax de Man, 1905, in Japanese waters is confirmed on the basis of five specimens from Iriomote Island, southern Ryukyu Islands. The species has been recorded with certainty only from Indonesia and New Caledonia. Upogebia pugnax is morphologically very similar to U. sakaii Ngoc-Ho, 1994, presently known only from Japan, with which it was previously confounded. Differentiating characters between the two species are reassessed. A brief overview on the Japanese species of Upogebia is given.
八重山諸島西表島において採集された標本5個体に基づき,アナジャコ科アナジャコ属のUpogebia pugnax de Man, 1905(新称:ヒメコブシアナジャコ)を報告した.本種名による日本からの記録はこれまでにもなされていたが,それらはコブシアナジャコU. sakaii Ngoc-Ho, 1994に帰されるもので,今回の記録により本種の日本沿岸での分布が初めて確認された.本種はU. sakaiiと混同されていたことが示すように,後者に形態的に酷似するが,口前部の棘の有無,尾節や尾肢外肢外縁の形状などで識別されることが指摘されていた.今回,識別形質の再評価を行い,これらの特徴に加え,第3および第4胸脚指節の屈曲縁に並ぶ櫛状剛毛の数と尾肢外肢のプロポーションにも種間で違いがあることを指摘した.さらに,本種は潮下帯に生息するが,コブシアナジャコは河口域や干潟域に主に生息するなどの生息環境の違いについても言及した.また,日本産アナジャコ属について簡単な分類学的概要を与え,現在のところ13種が記録されていることを示した.

概要: Approximately one-tenth of the 10 million individuals living with human T-cell leukemia virus type-1 (HTLV-1) worldwide live in Japan. Most of these infected individuals live in the southwest region of Japan, including Okinawa prefecture; however, currently no prophylactic vaccine against HTLV-1 infection is available. For preventing the HTLV-1 spread, we previously generated a humanized monoclonal antibody (hu-LAT-27) that mediates both neutralization and antibody-dependent cellular cytotoxicity (ADCC). The neutralization epitope of LAT-27 is a linear amino acid sequence from residue 191 to 196 (Leu-Pro-His-Ser-Asn-Leu) of the HTLV-1 envelope gp46 protein. Here, we found that the LAT-27 epitope is well conserved among HTLV-1 clinical isolates prevalent in Okinawa. The hu-LAT-27 treatment inhibited syncytium formation by these clinical HTLV-1 isolates. Although an amino acid substitution at residue 192 in the LAT-27 epitope from proline to serine was found in a few HTLV-1 isolates, hu-LAT-27 could still react with a synthetic peptide carrying this amino acid substitution. These findings demonstrate the wide spectrum of hu-LAT-27 reactivity, suggesting that hu-LAT-27 may be a candidate drug for prophylactic passive immunization against HTLV-1 infection.

概要: Numerous disease-causing gene mutations have been identified in proteinuric diseases, such as nephrotic syndrome and glo merulosclerosis. This report describes the results of comprehensive genetic diagnosis of Japanese patients with severe proteinuria. In addition, the report describes the clinical characteristics of patients with monogenic disease-causing mutations. We conducted comprehensive gene screening of patients who had either congenital nephrotic syndrome, infantile nephrotic syndrome, steroid-resistant nephrotic syndrome, or focal segmental glomerular sclerosis. Using targeted next-generation sequencing, 60 podocyte-related genes were screened in 230 unrelated patients with proteinuria. A retrospective review of clinical data was conducted for these patients. We detected monogenic disease-causing mutations in 30% (69 of 230) of patients among 19 of the screened genes. Common genes with disease-causing mutations were WT1 (25%), NPHS1 (12%), INF2 (12%), TRPC6 (10%), and LAMB2 (9%). With various immunosuppressive or renoprotective therapies, remission of proteinuria in patients with unknown causative mutations was observed in 26% of patients, whereas only 5% of patients with monogenic disease-causing mutations exhibited complete remission. We assessed the genetic backgrounds of Japanese patients with severe proteinuria. The proportion of patients with gene defects was similar to that of other reports, but the disease-causing gene mutation frequency was considerably different.

概要: Aim: This systematic review and meta-analysis evaluated whether bright light therapy (BLT) is an effective and safe trea tment for manic/depressive symptoms and a preventive measure for recurrent mood episodes in patients with bipolar disorder. Methods: A literature search of major electronic databases was conducted in June 2019, including all published articles up to that date. Two researchers independently selected relevant publications, extracted data, and evaluated methodological quality according to the Cochrane criteria. Results: Six randomized controlled trials (RCT) evaluated the efficacy of BLT for bipolar depression. A meta-analysis found no significant differences between BLT and placebo for the following outcomes: (i) rates of remission from depressive episodes (risk ratio [RR]: 1.81, 95% confidence interval [CI]: 0.43 to 7.64, P = 0.42); (ii) depressive symptom scores (standardized mean difference: −0.25, 95%CI: −0.74 to 0.23, P = 0.30); and (iii) rates of manic switching (RR: 1.00, 95%CI: 0.28 to 3.59, P = 0.26). The sensitivity analysis for studies with low overall indirectness did show a significant antidepressant effect for BLT (RR: 3.09, 95%CI: 1.62 to 5.90, P = 0.006). No RCT investigated the effect of BLT in preventing the recurrence of mood episodes in the euthymic state or in improving manic symptoms in the manic state. No severe adverse events were reported. Conclusion: While a meta-analysis was unable to demonstrate the efficacy of BLT for bipolar depression, a sensitivity analysis did show a significant effect. Further well-designed studies are needed to clarify the effectiveness of BLT, not only for the depressive state but also for other states, in the treatment of bipolar disorder. 続きを見る
